The widespread distribution in vivo and versatility of glutathione has attracted more and more attention of researchers. The glutathione plays a direct or indirect role in many significant organism phenomena and therefore involves various research topics such as enzymatic reaction mechanisms, biosynthesis of macromolecules such as proteins and DNA, intermediate metabolism, radiation, tumor, immunization, environmental toxins and aging, etc. In particular, many research topics have been involved in the defense of reactive oxygen species and free radicals in recent years. For the determination of biological samples, especially the concentration change of GSH and GSSG in serum, plasma is an important indicator of the body oxide.

Glutathione is a tripeptide (y-L-glutamyl-L-Cysteinyl glycine) and the most common intracellular thiol. In cells, glutathione exists in two different forms, reduced (GSH) and oxidized (GSSG). Under physiological conditions, more than 98% of intracellular glutathione is GSH, because GSSG is rapidly reduced to GSH by glutathione reductase. GSH-GSSG system is the most abundant oxidation-reduction system in eukaryotic cells. It plays an important role in cell homeostasis and participates in apoptosis-related signal transduction.

The Total Glutathione/Oxidized Glutathione BioAssay™ Kit measures glutathione (T-GSH) and oxidized glutathione (GSSG) in serum, plasma, animal tissue, whole blood, red blood cells and cultured cells.

Detection Principle:
GSSG is reduced to GSH by glutathione reductase and GSH can react with DTNB to produce GSSG and yellow TNB. The amount of total glutathione (GSSG+GSH) determines the amount of yellow TNB. Thus the total glutathione can be calculated by measuring the OD value at 412nm. The content of GSSG can be determined by first removing GSH from the sample with appropriate reagent and then using the above reaction principle.

Storage and Stability:
Store *405828B, *405828D, *405828H and *405828K at -20°C. Store all the other components at 4°C. Unused kit is stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vials after thawing and prior to removing the cap.
