-20°CWater is a critical component of all living cells. Interestingly, tissue membranes show a great degree of water permeability. Mammalian red cells, renal proximal tubules, and descending thin limb of Henle are extraordinarily permeable to water. Water crosses hydrophobic plasma membranes either by simple diffusion or through a facilitative transport mechanism mediated by special protein “aquaporin”. Over the last decade, genes for several members of aquaporin family (AQP1-9 and AQPQP) have been cloned, expressed, and their distribution studied in many tissues.|AQP2 or AQP-CD or WCH-CD (mouse, rat, human 271aa; human gene locus 12q13) is the vasopressin-regulated water channel of the apical membrane of collecting duct cells. AQP families of proteins are predicted to contain six transmembrane domains. The N and C-termini are predicted to be cytoplasmic. Defects in AQP2 are a cause of an autosomal dominant form of nephrogenic diabetes insipidus (NDI).||Applications: |Suitable for use in ELISA and Western Blot. Other applications not tested.||Recommended Dilution:|Western Blot: 1-10ug/ml using ECL. AQP2 is ~30kD (3). |ELISA: 0.5-1ug/ml. Conrtol peptide can be used to coat ELISA plates at 1ug/ml.|Optimal dilutions to be determined by the researcher.||Recommended Control Peptide: |A3000-07D: Aquaporin 2, Mouse, Control Peptide (AQP-CD, WCH-CD)||Storage and Stability:|Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. ||Note: Applications are based on unconjugated antibody.
