Deoxyribonuclease I, or DNase I

  • is used to nick DNA as a first step to incorporate labelled bases into DNA
  • is suitable for DNase footprinting
  • improves yield in primary cell isolation by removing extracellular DNA
  • reduces viscosity in protein samples by removal of DNA contamination

    DNase I is an endonuclease that cleaves phosphodiester bonds adjacent to pyrimidines to produce polynucleotides with terminal 5′-phosphates. It preferentially cleaves double-stranded DNA and, in the presence of Mg2+ , single-stranded DNA.



    Unit definition: 1 Kunitz unit catalyses an increase in absorbance of 0.001 at 260 nm per minute at 25 °C, pH 5.0, when exposed to highly polymerised calf thymus DNA (1).
