4°CIn situ hybridization (ISH) is a powerful method for gene expression analysis. This In situ Hybridization Kit (ISH), BioAssay™ provides the reagents, including hybridization buffer, NBT/BCIP color development solution and washing buffer solutions, for pre-hybridization, hybridization, and post hybridization treatments. This ISH kit provides researchers a method doing in situ hybridization with non-radioactive probes. Compared with radioactive methods, the sensitivity of this kit is the same while the procedure is simplified. This kit is suitable for digoxigenin labled probes combined with an alkaline phosphatase color development system. Using this kit, all disadvantages of working with radioactive materials can be avoided.

I7643-50C: NBT, 1x120ul or 1x300ul
I7643-50D: BCIP, 1x60ul or 1x150ul
I7643-50E: SSC Buffer, 20X, 1x20ml or 1x50ml
I7643-50F: Alkaline Phosphatase Buffer, 10X, 1x20ml or 1x50ml
I7643-50G: anti-Dig Antibody, 1x10ul or 1x25ul
I7643-50H: Blocking Solution, 100X, 1x100ul or 1x250ul
Buffer Required but Not Included:
1X PBS solution, pH 7.4: 81ml 0.2M Na2HPO4, 19ml 0.2M NaH2PO4, 18g sodium chloride
Add DEPC-treated water to 2000ml.
Storage and Stability:
Store components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Quality Control:
In order to ensure the quality of In situ Hybridization Kit (ISH), BioAssay™, a pre-test procedure is performed for each lot of the kits. The positive result is detected with a digoxigenin labeled Has-mirRNA 122a cDNA probe on a previous positive frozen tissue panel slide.
