Connexin 46 (Cx46), also known as Gap Junction Protein Alpha-3 (GJA3), CAE3, CX46 and C2P3, maps to human chromosome 13q11-q12 and encodes a 46kD protein. Cx46, along with Cx50, is principally expressed in the lens of the eye. Cx46 mediates intercellular interactions during development and is necessary for the survival of neural crest cells. Cx46 forms gap junctions that connect lens fiber cells and are crucial for maintaining lens transparency and normal lens function. Mutations of Cx46 result in severe cataracts of the lens.||Applications:|Suitable for use in Western Blot, Immunofluorescence and Immunohistochemistry. Other applications have not been tested.||Recommended Dilutions:|Western Blot: 1-3ug/ml. Identifies a single band at ~46kD|Immunofluorescence: 3ug/ml|Immunohistochemistry (Frozen): 3ug/ml|Note: For IHC, mice were perfused transcardially with 3ml pre-fixative (50mM phosphate buffer, 0.1% sodium nitrite, 1unit/ml heparin), followed by 40ml fixative (0.16M sodium phosphate buffer, pH 7.6, 0.2% picric acid, and 1-, 2-, or 4% paraformaldehyde), and perfusion with 10ml sucrose wash (10% sucrose in 25mM sodium phosphate buffer, pH 7.4. Tissues were then stored in cryoprotectant (10% sucrose, 25mM phosphate buffer, pH 7.4, 0.04% sodium azide) for a minimum of 24 hours prior to sectioning.|Optimal dilutions to be determined by the researcher.||Storage and Stability:|Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. |For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. ||Note: Applications are based on unconjugated antibody.
