Interferon-Gamma (IFN-γ) is a pleiotropic cytokine and the only member of the type II IFN family, which differs structurally and functionally from other types of IFNs. IFN-γ is a homodimer synthesized by non-covalent association of two 146 amino acid subunits in an antiparallel manner, forming a mature protein of approximately 33 kDa. Antigens, mitogens and cytokines, e.g. IL-12, IL-15 and IL-18, induce production of IFN-γ by T lymphocytes and natural killer cells. Once released, it acts as macrophage activation factor through binding of IFN-γ receptor I and II and subsequent activation of the JAK/STAT pathway. The inhibitory effects of IFN-γ on viral replication were originally reported in 1965 and its role in host response to microorganisms has been well established. IFN-γ also plays an important role in cancer immune surveillance, promoting tumor recognition and immunity.

Figure 2: Western blot analysis of PtX™ Mouse Anti-IFN-Gamma. Lanes 2–5: Varying amounts of IFN-γ were run on the SDS-PAGE. Separated bands were transferred to the membrane and PtX™ Mouse Anti-IFN- Gamma (1: 2 500) was used to detect the antigen. The two bands of IFN-Gamma were visualized following the addition of anti-rabbit secondary antibody with HRP and substrate. Our antibody was able to detect antigen amounts upwards of 31 ng (Lane 5).
